GC-MS Analysis of Free Amino Acids

Free amino acids (AAs) were extracted, derivatized and quantitated according to previously published method [37 (link)], with minor modifications. In brief, each sample (1g of fresh weight) was extracted with 15 mL of methanol and H₂O (7:3, v/v). The extracts were derivatized using an EZ:faast kit (Phenomenex, Torrance, CA, USA). The prepared samples were analyzed on a Hewlett Packard 6890N/5975 MSD gas chromatography-mass spectrometry (GC-MS) system (Agilent technologies).
