Before each test, the rats were placed in a Plexiglas box on the mesh grid for a 30 min acclimation period. Mechanical thresholds in rats were tested with von Frey probes (Stoelting, United States) as previously reported (Xu et al., 2018 (link)). Paw withdrawal mechanical threshold (PWMT) was recorded as the force of von Frey filament which induced at least three positive responses out of five applications. Thermal sensitivity thresholds were tested by Hargreaves Tes7370 (Ugo Basile, Italy) (Ding et al., 2017 (link)). Paw withdrawal thermal latency (PWTL) was recorded as the duration of radiant heat stimulation which was turned off automatically by paw withdrawal (cutoff time was 30 s to avoid tissue damage). PWMT and PTWL were measured in triplicate and averaged with an interval of 5 min. The motor function of rats receiving TSA or lentivirus injection was tested at day 10 or 14 after CCI, respectively, using rotarod test following the protocol we have described previously (Dai et al., 2019 (link)).
