Variable analysis
- Ground leaf material (100 mg)
- Soluble sugars
- Extraction in 1 mL of ddH2O by boiling for 15 min at 95 °C
- Vortexing and centrifugation for 10 min at 15,000× g
- Applying 200 µL of supernatant to Dowex® anion and cation exchange column
- Rinsing columns 6 times with 200 µL of ddH2O
- Dilution of flow through 1:1 in 20 µM rhamnose H2O as the internal standard
- Analysis by HPAEC-IPAD Dionex 5000 with separation on a CarboPacTM PA100 column and a mobile phase of 90 mM NaOH
- 10 µM of each sugar standard run alongside the samples
