Isolation of Lamellipodia Proteins

For proteins localized in the lamellipodia, cells were plated on 3.0 -µm porous polycarbonate Transwell membrane inserts (Costar^®) that were coated on the bottom with 25 µg/mL rat tail collagen (11179179001 from Roche^®). The lower chamber contained medium with 10% FBS, while the upper chamber was serum free. Cells were allowed to extend their lamellipodia through the pores. Cell bodies remaining on the upper surface were removed by scraping and the lamellipodia extending to the lower surface were recovered in lysis buffer^{13 (link),59 (link)}.
