Endothelial Progenitor Cell RNA Sequencing

Endothelial progenitor cells (EPCs) were harvested for total RNA extraction using Trizol® Reagent (Invitrogen). Extracted RNAs were measured the OD260/OD280 absorbance ratio to evaluate RNA quality by ND‐1000 spectrophotometer (Nanodrop Technology). Small RNA sequencing was performed by Welgene Biotechnology Company at the Solexa platform with a single‐end sequencing method in 75 nucleotides read length. Differentially expressed miRNAs greater than 2.0‐fold changes between young and senescent cell groups were indicated with reads per million (RPM) > 10 (Figure 1A).²⁰
