Isolation and Characterization of Lipid A from GMMA
Variable analysis
- Mild acid hydrolysis with 1% acetic acid for 2 h at 100°C
- Lipid A precipitation from GMMA
- Centrifugation at 14,000 g for 15 min
- Resuspension of pellets in water and washing twice with water
- Drying of pellets overnight using a Speedvac
- Resuspension of pellets in chloroform-methanol 4:1
- Mixing with an equal volume of Super DHB (Sigma-Aldrich) solution in water/acetonitrile 1:1 (vol/vol)
- Loading 2 μL of the mixture to the target plate
- Analysis by Ultraflex MALDI-TOF (Bruker Daltonics) in reflectron ion-negative mode
- Inclusion of Peptide Calibration Standard (Bruker Daltonics) mixed with the Super DHB solution in each analysis
- Determination of m/z ratios by Flex Analysis software in comparison to the Peptide Standard
