were custom ordered from GenScript. Peptides (>95% purity) were reconstituted to 1 mg/ml in sterilized water.
Cyclized peptide sequences for CF0 and CFC1 (biotin-HQCHQEST-X-GRSRGRCGRSGS, where X=Arg or Cit respectively) are based on
identified targets within human filaggrin (10 (link)). Previously described REP-1 and CEP-1
peptide antigen targets from human α-enolase protein (22 (link)) were similarly adapted
for cyclization (biotin-HQCKIHA-X-EIFDS-X-GNPTVEC, where X=Arg or Cit respectively). All subjects and controls were tested for
serological reactivity to each peptide (Supplementary Methods ). CFC1,
CF0, CEP-1, and REP-1 peptides were also separately loaded onto fluorescently–labeled streptavidin (SA) to generate
reagents for antigen-specific B cell detection and capture (Supplementary
