Culturing Pancreatic Cancer Cell Lines

Four pancreatic cancer cell lines (PANC-1, BXPC-3, ASPC-1 and CFPAC-1) were purchased from the Cell Bank of the Chinese Academy of Sciences (Shanghai, China). PANC-1 and BXPC-3 were cultured in Dulbecco’s modified Eagle medium (Thermo Fisher Scientific, South America) containing 10% fetal bovine serum (Thermo Fisher Scientific). ASPC-1 was cultured in RPMI-1640 (Thermo Fisher Scientific) containing 10% fetal bovine serum (Thermo Fisher Scientific). CFPAC-1 was cultured in Dulbecco’s modified Eagle medium (Thermo Fisher Scientific) containing 10% fetal bovine serum (Thermo Fisher Scientific). All cell lines were cultured at 37 °C under 5% CO₂/95% air and revived every 3 to 4 months. HSF1 and second mitochondria-derived activator of caspase (SMAC)-specific siRNAs were prepared and used in transfections as described previously [15 (link)].
