Genetic Manipulation of Sox2 and Ctnnb1 in Mice

All mouse experiments were performed in accordance with protocols approved under the guidance of the Institutional Animal Care and Use Committee of the University of Pennsylvania. Sox2^CreERT2 (Stock No. 017593), Sox2^Flox (Stock No. 013093), and Rosa26R^EYFP (Stock No: 007903) mice have been previously described and were obtained from Jackson Laboratories (MA, USA)^{47 (link),48 (link)}. Ctnnb1^flox(ex3) mice were described previously^{49 (link)}. Experiments were performed on 6-12 week old mice that were maintained on a mixed CD1 and C57BL/6 background. For all conditions, both male and female mice were used. Tamoxifen (Sigma-Aldrich) was dissolved in corn oil to prepare a stock at a concentration of 20 mg/mL. For gene knockout experiments, a dose of 200 mg/kg Tamoxifen was administered by oral gavage for 3 consecutive days.
