Evaluating Drug Effects on HCC Spheroids

Spheroids from HCC cells were generated using LIPIDURE^®-COAT PLATE A-U96 (NOF Corporation, Japan) according to manufacturer's instruction. Briefly, 3 days after seeding the spheroids were formed and treated with drugs or vehicle for 4 or 8 days. The effects of the drugs were evaluated in terms of volume changes using the Nikon Eclipse E400 Microscope with digital Net camera. The volume of spheroids [D = (Dmax + Dmin)/2; V = 4/3π (D/2)3] was measured using SpheroidSizer, a MATLAB-based and open-source software application (26 (link)).
