Cell Lysis and Protein Quantification

The target cell samples were washed with phosphate-buffered saline (PBS) two times, and the cell number was counted using Countstar cell analysis system (Alit Biotech, Shanghai, China). A total of 1 × 10⁶ cells were re-suspended in 200 μl CHAPS lysis buffer (Merck-Millipore) and incubated on ice for 30 min. The cell lysate was then centrifuged at 12,000 ×g for 20 min at 4°C. The protein concentration was determined by using Pierce BCA protein assay kit (Thermo Fisher Scientific). The supernatant were then transferred into new sterile tubes and stored at −80°C.
