Evaluating BI2536 and TMZ Efficacy in Glioblastoma

The animal experiment was approved by the Animal Care Committee of the First Affiliated Hospital of Soochow University. Code of Ethics: The First Affiliated Hospital of Soochow University 2018 Code of Ethics No. 117. To evaluate whether BI2536 and TMZ inhibit tumor growth in vivo, U87 stem cells were used to establish a nude mouse model. Female BALB/c nude mice (4 weeks, 15–17 g) were purchased from the Shanghai Experimental Animal Center of the Chinese Academy of Sciences (Shanghai, China). In order to establish an intracranial model, 5 × 10⁴ U87 stem cells were stereotactically injected into mice. The mice were randomly divided into four groups with 6 mice in each group. BI2536 was dissolved in 30% PEG-400, 0.5% Tween-80, 5% propylene glycol, and TMZ was dissolved in 5% DMSO + 30% PEG300 + ddH2O. During the survival period, mice were injected intraperitoneally with PBS, BI2536 (50 mg/kg/day), TMZ (20 mg/kg/day) or BI2536 (50 mg/kg/day) combined with TMZ (20 mg/kg/day))period. Animal research is conducted in accordance with internationally recognized guidelines and national regulations. The brain was extracted and fixed in 10% formalin, and then embedded in paraffin for immunohistochemistry.
