Variable analysis
- Gradient of buffer A (0.8 M ammonium acetate, pH 3) and buffer B (80% acetonitrile; LC-MS grade) applied at a flow rate of 1 ml/min
- Separation of pyridylaminated (PA) oligomannosidic N-glycans from white beans (containing Man3-9GlcNAc2)
- Isomeric/isobaric comparison of wild-type glycans using RP-HPLC
- IonPac AS11 column (Dionex, Sunnyvale, USA; 4 × 250 mm, combined with a 4 × 50mm guard column)
- Ascentis Express RP-amide column (150 x 4.6 mm, 2.7 µm; Sigma Aldrich, calibrated in terms of glucose units with an oligoglucose standard)
- Shimadzu Nexera UPLC system
- Fluorescence measured at 320 nm (excitation) and 400 nm (emission) with a Shimadzu RF-20A XS detector
- A pool of pyridylaminated (PA) oligomannosidic N-glycans from white beans (containing Man3-9GlcNAc2) was used to calibrate the column and test efficiency of separation.

About PubCompare
Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.
We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.
However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
