Characterizing Hemolytic, Urease, and Gelatin Liquefaction Activities of Lactic Acid Bacteria

For measuring hemolytic activity, each strain of LAB was inoculated on BD
BBL^TM prepared plated medium (Trypticase^TM soy agar
with 5% sheep blood; Thermo Fisher Scientific, Waltham, MA, USA) and incubated
at 37°C for 48 h. After 48 h, the colonies on plates were characterized
to observe the hemolytic pattern (Fu et al.,
2022). To detect urease activity, each strain of LAB was inoculated
on Remel^TM urea agar base (Thermo Fisher Scientific) and incubated at
37°C for 48 h. After 48 h, the color changes of plates were monitored to
observe the urease activity (Christensen,
1946). For gelatin liquefaction test, each strain of LAB
(5.0×10⁶ CFU/mL) was cultured in MRS gelatin broth
containing 12% (w/v) of gelatin for 48 h at 37°C. After 48 h, the
cultured MRS gelatin broths were incubated at 4°C to monitor gelatin
liquefaction (Fugaban et al., 2022 (link)).
