Culturing Pancreatic Cancer Cell Lines

AsPC1 & MIA PaCa2 were purchased from American Type Culture Collection (ATCC, Manassas, VA, USA), authenticated by STR profiling, and cultured as instructed by the vendor. Pan02 was purchased from ATCC. All culture materials were purchased from Gibco/Life Technology Inc., (Gibco, Grand Island, NY, USA). MMC18 cells, provided by Dr. Gloria Su, were generated from metastatic tumors of mouse pancreatic cancer model [17 (link)], and cultured in Dulbecco’s Modified Eagle’s Medium (DMEM) with 10% fetal bovine serum (FBS). AsPC1 was cultured in RPMI1640 Medium supplemented with 10% heat-inactivated fetal bovine serum, 100 U/mL penicillin and 100 mg/mL streptomycin. Other cell lines were cultured in Dulbecco’s Modified Eagle’s Medium (DMEM) supplemented with 10% heat-inactivated fetal bovine serum (Gibco, Grand Island, NY, USA), 100 U/mL penicillin and 100 mg/mL streptomycin. The cultures maintained in a humidified incubator at 37 °C and 5% CO₂.
