Ovariectomized Mice Treated with Tamoxifen

Mice were treated with Tamoxifen (2 mg/kg/day) using a regimen that we have previously shown synergistically enhanced loading-related bone gain [28 (link)]. At 16 weeks of age (day 1), 16 virgin female C57BL/6 mice were OVX. Ten days after surgery (day 11), the OVX mice were randomly subdivided into two groups (n = 8) and received either vehicle (peanut oil, 5 ml/kg; Sigma) or tamoxifen citrate (Tocris Cookson Inc., Ellisville, MO) by s.c. injection on days 11, 13, 15, 18, and 21 and were then sacrificed on day 25. Tibias were immediately snap frozen in liquid nitrogen and later used for qRT-PCR.
