In the initial approach, a targeted metabolomics approach using Waters’ ACQUITY UPLC® System with Xevo triple-quadrupole mass spectrometer (Waters, MA, USA) combined with the commercially available AbsoluteIDQ® p180 Kit (Biocrates Life Sciences AG, Austria) was used for rapid and quantitative analyses of 40 acylcarnitines according to the manufacturer’s protocol. The method combines the extraction of analytes with stable isotope dilution mass spectrometric analysis. The AbsoluteIDQ p180 Kit contains a 96-deep-well plate with a filter plate attached with sealing tape, as well as isotope-labeled internal standards and seven calibrators sufficient for quantitation of metabolites and three different quality control samples (low, medium and high spiked human plasma).
