Treg Cell Breeding and Characterization

Male mice of various ages were produced in our specific-pathogen-free facilities at Harvard Medical School, and were fed a chow diet (no. 5058, Lab Diet Picolab Mouse Diet 20), or HFD (60 kcal% Fat, Research Diets, D12492i). B6.CD45.1⁺ mice were purchased from the Jackson Laboratory. Foxp3-GFP mice were obtained from Dr. V. Kuchroo. vTreg53 TCR-tg mice were generated based on one of the expanded VAT-Treg clones, as previously described (Li et al., 2018 (link)), and were crossed with Foxp3-GFP^KI/KI females. Tg⁺ Foxp3-GFP^KI/y male progeny were used for experiments. In all cases, the Tcra plus Tcrb transgene was maintained in the heterozygous state. All experiments were performed using littermate controls and following animal protocols approved by the HMS Institutional Animal Use and Care Committee (protocol IS00001257).
