Mitochondrial Membrane Potential Analysis

The JC-1 MitoScreen kit (BD Biosciences, San Diego, CA, USA) was used for the mitochondrial membrane potential (ΔΨ_m) analysis. The assay was performed using a flow cytometer (BD FACSCanto II, Becton Dickinson Biosciences Systems, San Jose, CA, USA). The Les-3331 and reference drug were used in 1 µM and 5 µM concentrations. After 24 h incubation with tested compounds, the assay was performed as described in the literature [51 (link)]. Analysis of the obtained results was performed using FACSDiva software (version 6.1.3, BD Biosciences Systems, San Jose, CA, USA). The equipment was calibrated with BD Cytometer Setup and Tracking Beads (BD Biosciences, San Diego, CA, USA).
