Cells were treated with/without 100 ng/ml human recombinant CCL2 (Peprotech cat no. 300-04) or 20 nM INCB3284 (Cayman Chemical cat no. 11963) in growth media containing 2.5% Matrigel. Images were captured at day 4 for hDCIS.01 cells and day 8 for SUM225 cells at 10× magnification, 4 fields/well using an EVOS FL Auto Imaging system. Spheroid size was quantified using Image J software.
