African green monkey kidney cells (Vero cells) obtained from American type culture collection (ATCC) (cat # CCL-81) were cultured in Dulbecco’s modified Eagle’s medium (DMEM, Invitrogen, Carlsbad, CA, USA) supplemented with 10% fetal bovine serum (FBS, Invitrogen) (DMEM-10% FBS) at 37 °C with 5% CO2.
HSV-2 strain G obtained from ATCC (cat # VR-734) (KU310668) was propagated in Vero cells. Virus titration was performed by endpoint dilution and plaque assays.
ABMA [1-adamantyl (5-bromo-2-methoxybenzyl) amine] (Figure 1) was synthesized in-house. The positive control drugs, chloroquine and acyclovir, were purchased from Meilun Biotech Co., Ltd. (Dalian, China) [19 (link),20 (link),21 (link),22 (link),23 (link),24 (link)]. The purities of the compounds were higher than 98%, as determined by High Performance Liquid Chromatography (HPLC). The compounds were dissolved in dimethyl sulfoxide (DMSO) as stock solutions.
Specific-pathogen-free female BALB/c mice (6–8 weeks old) were obtained from the Changchun Institute of Biological Products and maintained under the guidelines for animal experiments at Jilin University, China.
