- Time duration of tissue digestion (60 min)
- Viable cell count
- Tumor tissue collection time (1-2 h after surgical removal)
- Washing of tissue in sterile Dulbecco's phosphate-buffered saline (PBS)
- Mechanical mincing of tissue into small pieces (2-4 mm)
- Digestion of minced samples using a tumor dissociation kit in a disposable gentle MACS™ C-Tube
- Digestion temperature (37°C)
- Filtration of digested samples through 70-μm sterile cell strainers
- Centrifugation of samples at 300 × g for 5 min
- Resuspension of cells in DMEM and HAM'S F12 media mixture (2:1) containing 50 IU/mL penicillin-streptomycin and 4 mM glutamine
