Xenograft Establishment in NSG Mice

NOD.Cg-Prkdc^scid Il2rg^tm1wjl /SzJ mice (NSG mice) were obtained from The Jackson Laboratory (Bar Harbor, ME, USA). All animal experiments were approved by the Seoul National University Institutional Animal Care and Use Committee (IACUC, SNU 15112–3-4). A total of 10 female NSG mice were used. Orthotropic xenografts were established via injection of 1 × 10⁶ shNSDHL MDA-MB-231 cells (n = 5) or shControl cells (n = 5) mixed with Matrigel (BD Biosciences) into the fat pad of the 4th mammary gland of 5-week old mice. After injection of tumor cells, primary tumor volume was measured weekly using digital calipers and a modified ellipsoidal formula (volume = 1/2(length×width²)) [17 (link)].
