FTIR Analysis of MoPrP Fibrils

Variable analysis
- Dialysis of MoPrP fibril samples into MilliQ water for 24 hours at 4 °C
- Centrifugation of fibrils at 20,000 x g for 30 min to separate them from buffer solution
- Resuspension of fibrils in 1 mL of D2O, repeated 3 times
- Sonication of all samples for 60 s (MS-72 tip, 20% sonication strength)
- FTIR spectra recorded using Bruker Vertex 80 v IR spectrometer equipped with MCT detector
- Use of CaF2 transmission windows and 0.05 mm Teflon spacers for all measurements
- Measurements performed at room temperature under near-vacuum conditions (~2 mBar)
- Subtraction of D2O spectrum from each sample spectrum
- Normalization of all spectra to the same area of amide I/I' band (1700–1595 cm^-1)
- Data processing using GRAMS software
