Culturing Mycobacterium tuberculosis H37Rv

M. tuberculosis H37Rv (ATCC 27294) with (H37Rv:mCHERRY) or without (H37Rv):pCHERRY3 reporter plasmid [22 (link)] was cultured in Middlebrook 7H9 liquid media (Becton Dickinson, USA) supplemented with 0.2% (v/v) glycerol (Merck Laboratories, USA), 0.05% Tween 80 (Sigma-Aldrich, Germany) and 10% albumin-dextrose-catalase (ADC) (Becton Dickinson, USA), in filtered screw cap tissue culture flasks (Greiner Bio-one, Germany). Hygromycin B (50μg/ml) was included for plasmid maintenance, where required. The cultures were incubated at 37°C until an optical density (OD₆₀₀ = 1.0) was reached.
