Quantifying Genomic 5-mdC by cHILIC-ESI-MS/MS

The capillary hydrophilic interaction chromatography (cHILIC) was performed on a Shimadzu Prominence nano-flow liquid chromatography system (Shimadzu, Tokyo, Japan) with two LC-20AD nano pumps, two vacuum degassers, a LC-20AB high performance liquid chromatography (HPLC) pump, a SIL-20AC HT auto-sampler, and a nano-flow control valve. The electrospray ionization/tandem mass spectrometry (ESI-MS/MS) experiment for detecting the genomic 5-mdC contents was detailly described in the previous study [29 (link)]. The results showed linearity within the range of 0.05% - 10% (molar ratio of 5-mdC/dC) with a coefficient value (R²) 0.996.
