Liver Injury Histological and Molecular Assessment

Blood was collected from the IVC, and the livers were recovered. Serum alanine transaminase (ALT) was measured using analytical kits from Pointe Scientific (Uncoln Park, MI) [8 (link)]. Histology was performed using hematoxylin-eosin (H&E) staining. Necrotic areas were quantified in a blinded manner by image analysis of 10 randomly selected fields per liver in slides stained with H&E using a IPlab 3.7v software (BD Biosciences, Rockville, MD) [9 (link)]. Apoptosis was assessed by terminal deoxynucleotidyl transferase dUTP nick-end labeling (TUNEL) using an In Situ Cell Death Detection Kit [10 (link)]. RT-PCR was used to determine the expression of transforming growth factor-β (TGF-β) mRNA.
