Evaluating Cordycepin Cytotoxicity in MEFs

Cordycepin (Sigma-Aldrich) was dissolved in DMSO (Sigma-Aldrich) and stored at −20 °C. The cytotoxicity of Cordycepin in MEFs was assessed following previously described MTT procedures^{15 (link)}. Briefly, cells were treated with different concentrations of Cordycepin (DMSO: 0.1%) for different time interval before four-hour incubation with medium containing MTT (3-(4,5-dimethylthiazol-2-yl)−2,5-diphenyltetrazolium bromide; Sigma-Aldrich). After incubation, the purple crystal (formazan) was dissolve in DMSO and the absorbance was read at 540 nm with a standard plate reader.
