Sodium Arsenite Lethality Assay

Using 96-well plates, day 3 adult worms were transferred to wells containing 100 µL of sodium arsenite (Sigma-Aldrich, S7400) solution in concentrations ranging from 10 to 15 mM, diluted in M9 buffer (22 mM Na₂HPO₄, 22 mM KH₂PO₄, 85 mM NaCl, 1 mM MgSO₄). After 8 h, dead worms were scored by touching their tail three times with a platinum wire. When touch response was absent, worms were considered dead. At least 6 wells containing 10 worms were used for each strain and each concentration. The experiments were repeated at least 2 times.
