LSCC Cell Line Characterization and Inhibition

The human LSCC cell lines of AMC-HN-8 and TU177 were purchased from Beijing Beina Chuanglian Institute of Biotechnology (Beijing, China). The AMC-HN-8 cells were cultured in Dulbecco’s modified Eagle’s medium (DMEM), containing 10% foetal bovine serum (FBS). The TU177 cells were cultured in RPMI-1640 medium, supplemented with 10% FBS. All of the above cell culture reagents were provided by GIBCO (Thermo Fisher Scientific, Inc). Accordingly, the culture of two cells was performed in a humidified incubator with 5% CO₂ atmosphere at 37°C (Thermo Fisher Scientific, Inc). The PI3K inhibitor (LY294002) and mTOR inhibitor (Rapamycin) were purchased from MCE (New Jersey, USA). LSCC cells were incubated with LY294002 or Rapamycin to perform functional assays and Western blot assay.
