MTT Assay for Cytotoxicity Evaluation

Cell cytotoxicity was determined with the MTT based-In Vitro Toxicology Assay Kit. Cancer cells were treated with cottonseed extracts and incubated at 37 °C, 5% CO₂ for 2 and 24 h. The cell media were added with 50 µl of MTT assay reagent (thiazolyl blue tetrazolium bromide) and incubated at 37 °C, 5% CO₂ for 2 h before adding 500 µl MTT solubilization solution to each well, shaken at room temperature overnight. The color density in the wells was recorded by Epoch microplate spectrophotometer at A570 after diluting 8 times.
