Variable analysis
- Cell lysis with Laemmli sample buffer containing 2.5% β-mercaptoethanol
- Heating at 95°C for 5 min
- Protein expression levels of MMP14, alpha-catenin, Vimentin, Fibronectin, integrin β1, integrin β3, and actin
- 4–15% polyacrylamide gels (Bio-Rad) for electrophoresis
- Protein transfer to Poly Vinylidene DiFluoride (PVDF) membrane (Merck)
- Blocking with 5% dry milk, Tris buffered saline, 0.2% Tween
- Incubation with primary antibodies (overnight at 4°C)
- Incubation with secondary antibodies (1:10000) for 1 hr at room temperature
- Protein detection using Luminata Crescendo (Merck) and LAS600 (GE Healthcare)
