Charcoal Meal Transit Assay in Rats

The gastrointestinal motility was measured by the method described by Choi et al. [51 (link)] and Kim et al. [52 (link)] with some modifications. Briefly, SD rats were fasted for 18 h prior to the experiment but were allowed to consume water ad libitum. These rats were fed with 1 mL of charcoal meal (3% suspension of activated charcoal in 0.5% aqueous methylcellulose) (Sigma-Aldrich Co., St. Louis, MO, USA). After 30 min of treatment, these rats were euthanized using CO₂ and the intestinal tract was collected from the abdominal cavity. The intestinal charcoal transit ratio was calculated as follows:
