Evaluating NS MIX Cytotoxicity in Vero Cells

The cell viability of samples treated with NS MIX was measured by using the ViaLightTM plus cell proliferation and cytotoxicity bioassay kit, according to the manufacturer’s instructions (Lonza Group Ltd., Basel, Switzerland). Vero cells were grown in 96-well plates and treated with NS MIX at different concentrations (1.6, 0.8, 0.6, 0.4, 0.2, and 0.1 mg/mL). DMSO was used as negative control. After 72 h the ATP levels was quantified in a GloMax Multi Microplate Luminometer (Promega Corporation, 2800 Woods Hollow Road Madison, WI, USA). The luminescence value was converted into the cell proliferation index (%), as previously reported [11 (link)]. The 50% cytotoxic concentration (CC₅₀) was calculated from concentration-effect curves by using non-linear regression analysis. The GraphPad Prism 6 software was used for data analysis. The results are the means (± SD) of three independent experiments.
