Fluorescent Labeling of Degenerating Neurons

Fluoro-Jade B (FJB; Chemicon, Temecula, CA, USA) is a polyanionic fluorescein derivative that labels degenerating neurons with high sensitivity and specificity [33 (link)]. Sections were rehydrated in graded ethanol solutions (100% and 70% for 5 min each) and distilled water, incubated in 0.06% KMnO₄ followed by a 0.001% solution of FJB for 30 min each, and observed under a fluorescence microscope (Olympus BX-51; Olympus, Tokyo, Japan) at 450–490 nm.
